Mushrooms can only grow in the Caves when using a Farming Frame. They can also be found naturally in the Caves and Dungeons as well and are illuminated in these dark areas to be easily discovered. Mushrooms make a great simple hunger boost but can also be used in cooking recipes to make high-tier food using a Fireplace.
